Hello!
EE’s support license can only be used once, but I would like to use the plug-ins on a development/test website to show a prototype to the client and to have everything ready to go live when it is done.
Can I later request a transfer of the license to the new site/URL when it is live?

You could request a key reset later, but you don’t need a support license key for your website that you’re showing as a prototype. Everything will work without the license key, except for one-click updates. You probably don’t need 1-click updates for the prototype site right?

Do all the plug-ins work without the support license? That was the only thing I was looking to use.

Yeah there are no restrictions with the plugin functionality when it comes to the license keys. The only difference will be the demo site won’t be able to do 1-click updates without the key.
